ABSTRACT:
This study optimized probe sonication-assisted extraction of polyphenols from Terminalia bellerica, Kigelia pinnata, and Aerva lanata using Box-Behnken Design (BBD) and Response Surface Methodology (RSM). The effects of solvent composition (0-100% ethanol), sonication time (5-60min), and amplitude (10-98%) on total phenolic content (TPC) were evaluated. RSM analysis revealed optimal conditions: T. bellerica achieved maximum TPC (602.42±9.23µg GAE/mL) at 62.08% ethanol, 34.39min, and 62.37% amplitude; K. pinnata yielded 60.73±2.56µg GAE/mL at 44.26% ethanol, 27.03min, and 55.41% amplitude; and A. lanata showed highest TPC (34.24±1.28µg GAE/mL) at 46.54% ethanol, 20min, and 44.79% amplitude. The validated model confirmed probe sonication as an efficient, scalable method for polyphenol extraction, highlighting its potential for pharmaceutical and industrial applications.
