ABSTRACT:
Background: Honey, a multifaceted mixture of enzymes, peptides and amines, has domestic as well as medicinal utility. Recent studies that analyzed its effects in different strains of rat and mouse have pronounced honey as antimetastatic with moderate antitumor effects. In the present study we have tested the cytotoxic activity of diluted unfractionated bee honey against Saos-2 (Sarcoma osteogenic) cell line, a primary osteosarcoma in vitro, to demonstrate that honey can induce apoptosis in primary osteosarcoma cells. Methods: The cytotoxicity effect of honey was evaluated on SAO’S cells and cell cycle analysis was performed using propidium iodide. To look for DNA breaks in cells, DNA of the sample was run through electrophoresis in standard TE buffer to visualize DNA separation pattern on a UV Transilluminator. Results: Cell viability was steadily lost with increasing concentrations of honey and IC50 value for honey was found to be lower than methotrexate drug. Phase contrast microscopy showed that cell appearance was indicative of apoptosis. Results showed that Honey showed an arrest at G0/G1 phase. DNA fragmentation exhibited DNA fragmentation. Methotrexate in combi¬nation with honey resulted in the formation of a set of defined wreckage as characteristic of apoptosis. Discussion: The study indicates that honey can be considerably effective against cancer cell lines in vitro. Further in depth study would help to explain the exact mechanism of action of honey against cancer cells and other malignancies.
