ABSTRACT:
Introduction: Nisin is a bacteriocin that has antibacterial activity against both gram-positive and gram-negative bacteria. For the sake of potentially topical use, the release of nisin is expected to be slowed down.Microsphere is an attempt for slowing down the release in topical delivery system. Purpose: The purpose of this research is to find out the characteristic of nisin microspheres produced by ionotropic gelation method and aerosolization technique with sodium alginate-gelatin and sodium alginate. Methods: This research was conducted by using qualitative analysis through the collected raw materials. The writers used ionic gelation method and aerosolization technique toward polymers withsodium alginate-gelatin (2.25%: 0.25%) and compared to microspheres made by polymers withsodium alginate (2.5%). The techniques of data analysis used in this research were organoleptic, infrared spectroscopy, and X-ray diffraction. Results: The resultof morphological observation shows that the microspheres are in spherical shape with a smooth and flat surface. The observation of particle size using optical microscope shows the microsphere particles are produced in size between 2.861-7.439 µmin polymers withsodium alginate-gelatin (Formula I) and 3.004-9.871 µmin polymers withsodium alginate (Formula II). The average particle size based on the calculation obtains the number of 4.048 + 0.069 µm for Formula I and 5.656 + 0.130 µm for Formula II. Conclusion: The different characteristicbetween polymers nisin microspheres with sodium alginate-gelatin and polymers with sodium alginate is found in the average particle size. The number of average particle size of nisin microspheres with a combination matrix is smaller than the average particle size of nisin microspheres with sodium alginate matrix.
