ABSTRACT:
Pulmonary Arterial Hypertension (PAH) is a steadily advancing, enfeebling disorder marked by raised pulmonary arterial pressure and peripheral resistance, often leading to right-sided heart failure. A dual endothelin receptor antagonist Bosentan monohydrate is efficacious in managing PAH but faces limitations such as a short half-life of 5 hours, limited oral absorption, a narrow window of absorption in the upper gastrointestinal tract and hepatotoxicity. To address these challenges, a gastro-retentive floating drug delivery system of Bosentan Monohydrate was formulated using direct compression method. A 3² full factorial design was employed to optimize the formulation by assessing the effects of AnyCoat CN15U Plus (sustained-release polymer) and sodium bicarbonate (carbonating agent) on floating lag time and amount of drug release at 2, 8, and 14 hours. Among the batches, formulation F5, containing 80 mg of Sustained release polymer AnyCoat CN15U Plus and 40 mg of carbonating agent sodium bicarbonate, exhibited a floating lag time of 5 seconds and sustained release of approximately 90% over 14 hours. Pre-compression and post-compression parameters confirmed acceptable physicochemical properties.
