ABSTRACT:
A novel, safe, accurate and sensitive spectrophotometric method was developed using 2M Urea as hydrotropic solubilizing agent for the quantitative determination of poorly water soluble drug Olaparib in tablet dosage form. The results of the investigation revealed a significant improvement in solubility, showcasing more than a 21-fold enhancement compared to the solubility in distilled water. Olaparib shows maximum absorbance at 276 nm. Urea did not show any absorbance above 240nm and thus no interference in the estimation of drug was seen. Olaparib follows the Beer’s law in the concentration range of 10-20µg/mL (r2 = 0.9995). The average recovery at three different concentration levels were found in the range of 98 – 102%. Olaparib’s LOD and LOQ were determined to be 0.619µg/mL and 1.87µg/mL respectively. The mean percentage assay of olaparib in tablet dosage forms by proposed method were found to be 99.50±1.19. The accuracy, linearity, precision, robustness, ruggedness, LOD, and LOQ of the proposed method have been validated in accordance with ICH guidelines. The proposed method is novel, rapid, simple to follow, precise, and consistent. The accuracy, reproducibility, and precision of the proposed method has been demonstrated by statistical data. This study underscores the importance of hydrotropic agents in addressing solubility challenges and highlights their potential impact on analytical methodologies and drug development processes.
