ABSTRACT:
Asenapine Maleate (ASM) is atypical antipsychotic drug used in schizophrenia. Asenapine maleate is a BCS-II having less than 2% bioavailability due to extensive 1st pass metabolism. In this study, enhancement of low oral bioavailability of Asenapine maleate by Nano lipid carrier approach with thin film hydration technique. A single dose pharmacokinetic investigation in New Zealand healthy rabbits found a considerable rise in ASP concentration in the brain after intranasal administration of ASP-MNE, reaching a maximum of 997.3.±3.07ng h/ml. Bioanalytical procedures used to measure drug catabolites in biological fluids have significance in the assessment and interpretation of bioavailability. The development of selective and sensitive bioanalysis of analytes in order to conduct pharmacokinetic studies successfully. When compared to other analytical techniques, LC-MS detection is the greatest option for developing a method with the appropriate LLOQ and ULOQ, as well as accuracy. AM-SLN had significantly higher (P<0.05) Cmax (30.52±0.56ng/mL), AUC (997.3±3.07h ng/mL), and MRT (22.9±0.5 h) than AM-solution (21.93±1.21ng/mL, 97.67±1.34ng/mL, and 4.3±0.08 h, respectively) when taken orally. A study of brain distribution discovered that medicines passed across the blood-brain barrier more easily and accumulated in the brain. These findings imply that SLNs might be a unique and promising drug delivery technique for oral AM administration in the treatment of psychotic disorders.
