ABSTRACT:
The aim of this study was to evaluate the feasibility of obtaining prolonged-release solid dispersions of trimetazidine dihydrochloride using kollidon® SR and PolyoxTM WSR 303 in various ratios as drug release-slowing polymers. Solid dispersions in the ratios of 1:2, 1:4 and 1:8 (drug: polymer) were prepared by solvent evaporation method using isopropanol as solvent. The prepared formulations were evaluated for drug content uniformity, flowability, compressibility, weight loss on drying, and dissolution profile. Structural and morphological characteristics were assessed using X-ray diffraction and microscopy. Uniform distribution of the drug throughout the polymer matrix was demonstrated for all formulations, with drug content varying from 97.14 to 103.33%. The weight loss on drying was in the range of 1.12-2.84%, and flowability and compressibility were within the satisfactory range, varying from fair to excellent. Increasing the proportion of polymer reduced the dissolution rate, enabling prolongation of the drug release up to 12 h with Kollidon® SR. However, a prolonged-release profile could not be obtained in solid dispersions prepared with PolyoxTM WSR 303. X-ray diffraction confirmed the formation of solid dispersions and the absence of polymorphism, while microscopy provided morphological evidence of solid dispersion formation. The results of this study demonstrate that prolonged-release solid dispersions of the water-soluble drug trimetazidine dihydrochloride can be obtained by solvent evaporation method using appropriate polymers in suitable ratios.
