ABSTRACT:
Corchorus depressus exhibits several pharmacological activities, including aphrodisiac, antipyretic, and demulcent effects, and is used in the treatment of gonorrhea. This study aims to systematically standardize Corchorus depressus, focusing on evaluating its standardization parameters, as no comprehensive standardization has been reported to date. The markers Diosgenin and Stigmasterol in the root powder were quantified using a developed and validated HPLC method. Various standardization parameters were assessed, including organoleptic properties, macroscopy, powder microscopy, physicochemical parameters, preliminary phytochemical screening, microbial contamination, and toxicological profiles. The HPLC method that was developed turned out to be straightforward, linear, accurate, precise, and selective. Chromatographic separation was achieved on a Symmetry C18 column using isocratic elution and a mobile phase consisting of acetonitrile and water (98:2, v/v). At 210nm, a UV detector was employed for detection. Diosgenin and stigmasterol had retention periods of 5.6 and 10.2 minutes, respectively. After being validated in compliance with ICH guideline Q2(R1), the method showed satisfactory system suitability characteristics and analytical performance. The root powder of Corchorus depressus was discovered to contain 0.281% w/w of diosgenin and 0.312% w/w of stigmasterol. This approved approach allows for the simultaneous quantification of these phytoconstituents.
