ABSTRACT:
Ca2+/Al3+ -ion induced PCM+MET -loaded alginate-carboxy methyl alginate blend beads were successfully prepared using Ca2+/Al3+ -ion induced ionic-gelation technique in an aqueous medium. Effects of polymer-blend ratio and concentration of cross-linker on the entrapment efficiency (DEE) and drug release at 8 h (R8h) of PCM+MET were optimized using 32 factorials. Optimization of the formulation of Ca2+/Al3+ -ion induced PCM+MET loaded alginate-carboxy methyl alginate beads gave DEE between 30%-76%, with 100% drug release in 8 h time point. The bead size varied from 786.45±0.133µm to 1060±0.155µm. The bead surface morphology was determined using SEM. The drug-polymer interactions were confirmed in the optimized formulation using FTIR and p-xrd analysis. These beads released drugs in a prolonged period of time in an in vitro release study, and follow a zero-order release profile, with a super case-II diffusion mechanism. Swelling and degradation of the optimized beads are dependent on the pH levels of the medium tested, and could be utilized for a GRDDS.
