ABSTRACT:
In this study formulation and evaluation of ethosome-based topical gel formulations for enhanced transdermal supply of Dapsone and Cloxacillin Drug-excipient compatibility was confirmed by FTIR, indicating no drug-drug and drug-excipients interaction. Ethosomes (F1–F9) were categorized for vesicle size (110.2–176.7nm), zeta potential (–13.2 to –21.7mV), and high encapsulation efficiency (49.07±0.85 – 71.91±0.98), with optimized formulations (F1–F3) showing greater physicochemical properties. Among these, F3 is the best formulation in comparison to FI,F2 & F3. Ethosomal gels (G1–G9) were formulated using Carbopol 934 and calculate the drug content, spreadability, pH, and viscosity. Optimized gel G3 confirmed high drug content (77.15%), pH (7.2), and ideal spreadability. In-vitro and ex-vivo studies show sustained drug release (up to 89.45% over 24 h) and the ethosomal gel (G1-G3) have a significantly longer release (76.5±0.45, 81.5±0.33, 89.45±0.27%) in comparison to conventional gel, CG (26.09±1.17%) in 24hr. TEM and SEM show spherical morphology with even vesicle distribution. PEG 400 and low Carbopol concentration in G3 enhanced vesicle flexibility and skin permeation. Overall, Gel (G3) show as a stable, active transdermal delivery system for enhanced topical delivery of Dapsone and Cloxacilline. The existence of Dapsone and Cloxacilline in ethosomes is able to enhanced permeation. The studies show that the optimized vesicular gel demonstrated a 3.42 times higher maximum in comparison to the conventional gel.
